Adryl (Diphenhydramine Hydrochloride) Syrup
Indications: Adryl is indicated for the treatment of the following conditions:
- Seasonal, perennial, and vasomotor rhinitis
- Urticaria, angioneurotic edema, and anaphylaxis
- Pruritic conditions
- Premedication for emesis and motion sickness
- Miscellaneous conditions such as Meniere’s disease and parkinsonism
Pharmacology: Diphenhydramine, the active ingredient in Adryl, is an antihistamine with anticholinergic and sedative properties. It competes with histamine for H1-receptor sites on effector cells in the gastrointestinal tract, blood vessels, and respiratory tract, thereby alleviating allergic symptoms.
Dosage Guidelines:
For Adults:
- Allergic conditions: 25-50 mg, three times daily, with an additional 50 mg at bedtime if needed.
For Children:
- Ages 1-5: 5 mg (2.5 ml of syrup) four times daily.
- Ages 6 and above: 10 mg (5 ml of syrup) four times daily.
Route of Administration:
- Administered orally.
Interactions:
- Adryl reduces the absorption of para-aminosalicylic acid (PAS), an antituberculous agent, from the gastrointestinal tract.
- CNS depressants can enhance the sedative effects of Adryl.
- Anticholinergic drugs may amplify Adryl’s anticholinergic side effects.
Contraindications:
- Hypersensitivity to Diphenhydramine Hydrochloride.
- Ammonium chloride is contraindicated in patients with impaired hepatic or renal function.
Side Effects: Possible side effects include:
- Sedation, dizziness, tinnitus, fatigue, ataxia, blurred vision, diplopia, euphoria, and gastrointestinal discomfort.
Pregnancy & Lactation:
- Category B: There are no adequate and well-controlled studies in pregnant women. Diphenhydramine should only be used during pregnancy if clearly necessary.
- Diphenhydramine is excreted in breast milk, and its use in breastfeeding mothers is not recommended.
Precautions & Warnings:
- Exercise caution in patients where drowsiness could impair functioning, such as drivers or machine operators.
- Alcohol and CNS depressants may intensify drowsiness.
Therapeutic Class:

- Store at temperatures between 15-30°C and protect from moisture.
